The 25 cm column extends separation space beyond mid-length formats, directly increasing peak capacity and reducing co-elution across complex peptide mixtures.

At 400 nL/min, FWHM values of ~4.2–5.5 s across 30–60 min gradients provide the resolving power required for deeper identification and improved precursor separation. The increased column length enhances proteome depth without requiring extreme gradient durations.
Operating within 100–800 nL/min, the 25 cm format is optimized for high-resolution nanoflow separations where peak capacity is the primary driver of identification depth.

Core columns are bare fused-silica emitter-integrated analytical columns designed to be connected via a liquid junction. With an outer diameter of 363 μm, they are compatible with liquid junctions engineered for standard 360 μm fused-silica tubing. Depending on the instrument setup, metal junctions can provide grounding or high-voltage transfer, while PEEK junctions may be used when an external grounding or voltage supply point is available.
Core columns can also be mounted directly in nanospray sources that are designed to accept bare emitter tips. This includes Thermo Scientific Nanospray Flex and Bruker CaptiveSpray sources (with appropriate emitter positioning accessories such as the CaptiveSpray (CSI) emitter adjustment kit), enabling use of the column itself as the electrospray emitter and minimizing post-column volume.
Specific adapter configurations may be required depending on the vendor ion source when using bare emitter columns. Users should consult instrument documentation where compatibility with blank-emitter columns is explicitly described.
